Prometheus was a big hit at the box office in its opening weekend, bringing in over $50 million in ticket sales but the overall opinion of the movie seems to be split. Some really liked it while others, including many hardcore Alien fans, hated it.
Whether you liked it or not, it was a thought provoking film that left many questions unanswered which in turn has confused many who have gone and seen it already.
SPOILER ALERT. If you haven't seen this movie yet, and don't want to have anything ruined, please stop reading this now but please come back and read this opinion column after you've seen it.
At the beginning of Prometheus you see an alien humanoid who is called an "Engineer" being dropped off on a planet and standing along running water and a waterfall. As the alien watches a spaceship take off back into space, the lone humanoid drinks some black goo. After consuming it a reaction starts to take place and the alien looks surprised at the effect as his body breaks down and disintegrates. He falls into the water and down into the waterfall. As he settles at the bottom of the water, his DNA is shown to be breaking apart but then reconstructing itself into new cells/DNA.
The assumption here is that this is how life on Earth was created, by the Engineers.
Later in the movie, Prometheus' ship captain states how the alien establishment they're exploring is something of a military establishment where weapons testing could have been the primary objective of the settlement. However, he says something must have went terribly wrong and the Engineers had to abandon the moon and the weapon entirely.
This statement made this journalist start to connect some dots from the beginning of the movie. The Engineers weren't attempting to create new life on Earth but since it was an uninhabited planet, it made the perfect testing ground for their new chemical/biological weapon (the black goo). The test went bad and the unknown side effect was the creation of life on the previously unpopulated planet.
This is why, later in the movie, the viewer learns the Engineers wanted to go back and remove life from Earth, which was the accidental side effect of their chemical/biological weapon test when the cells/DNA were shown reconstructing themselves.
Maybe this isn't the meaning behind this concept in Prometheus but it would make the most sense and gives them reason for a sequel. Especially since Elizabeth Shaw and David are now off to find the Engineers and ask them why they wanted to destroy life on Earth after they "created it."
Again, this is only one person's theory but it's logical and puts more pieces of the puzzle together in order to find an answer to a question that is currently being asked by those who have seen the movie already.

(Note: For those who think the Engineers came back to Earth several times and that was the reason behind all the cave paintings, please understand that was never said in the film either and is as much of a theory as this opinion column is too. It is an assumed truth the Engineers came back and that is why the paintings were made but this was never actually said to have happened. We see these paintings as part of the genetic connection to the Engineers just like how the black goo pillars are arranged exactly like how the Alien Queen lays her eggs, due to the genetic connection from the originator.)
